Biosynthesis of Pyrrole-2-carbaldehyde via Enzymatic CO2 Fixation

The use of CO2 as a chemical building block is considerable interest. To achieve carbon fixation at ambient conditions, (de)carboxylase enzymes offer an attractive route but frequently require elevated [CO2] levels to yield the acid product. However, it has recently been shown that coupling UbiD-type decarboxylase with carboxylic reductase yields corresponding aldehyde product near [CO2]. Here, we show this approach can be expanded different UbiD and CAR alternative products, in case, production pyrrole-2-carbaldehyde from pyrrole, using Pseudomonas aeruginosa HudA/PA0254 combination Segniliparus rotundus CAR. This confirms varied substrate range respective harnessed distinct combinations support wide aldehydes via enzymatic fixation.

The title compound, C(7)H(7)NO(2), was synthesized via a one-pot Vilsmeier-Haack and subsequent Friedel-Crafts reaction. The pyrazole ring makes dihedral angles of 4.50 (9) and 2.06 (8)°, respectively, with the aldehyde and acetyl groups. In the crystal, classical N-H⋯O hydrogen bonds and weak C-H⋯O inter-actions assemble the mol-ecules into a chain along the b axis.
